Research Associate, Immuno-Oncology

Soteria Biotherapeutics is a San Mateo-based biotechnology company developing next-generation T-cell engager therapies. Our Immuno-Oncology group is looking for a Research Associate with experience performing cell-based assays to contribute to Soteria’s discovery pipeline. This is a hands-on role in which you will assess new drug candidates using a variety of liquid-handling instruments to execute cell-based cytotoxicity and flow cytometry assays. You will generate data on bleeding-edge molecules that informs the iterative process of antibody engineering. We are looking for candidates who are methodical, detail-oriented, eager to learn new skills, and excited about joining a growing biotech company.

Primary Responsibilities

  • Support in vitro experiments to prioritize and characterize new drug candidates
  • Report to a Senior Scientist, Immuno-Oncology and work with them to design and interpret experiments
  • Perform T-cell-dependent cytotoxicity co-culture assays using a semi-automated liquid-handling workflow
  • Measure antigen expression by flow cytometry and work with Scientists to optimize new flow assays
  • Analyze your data and present your results at team meetings
  • Operate instruments for flow cytometry, real-time microscopy, and plate-based measurements
  • Maintain sterile cultures of mammalian cell lines and primary immune cells
  • Isolate and cryopreserve human immune cells from leukapheresis packs
  • Prepare sterile growth media for tissue culture
  • Document experimental procedures and results in a digital lab notebook and inventory system
  • Proactively identify and execute ways to keep the lab running smoothly

Required Qualifications

  • BA or BS in biology, biotechnology, immunology, biochemistry, pharmacology or a related discipline
  • Demonstrated interest in cancer biology or immunology
  • 1+ years of experience performing cell-based assays (academic research included)
  • Experience performing tissue culture of mammalian cells
  • Strong communication and interpersonal skills
  • Track record of careful planning of experiments and documentation of results
  • Enthusiastic, self-motivated, and eager to contribute to a nimble, fast-paced team environment

Additional Preferred Experience

  • Performing CellTiter-Glo or ELISA assays
  • Using Cytek flow cytometers or IncuCyte real-time microscopes
  • Using plate-based liquid handlers (e.g. Wellmate, Assist, Bravo, Biomek, Opentrons, etc.)
  • Analyzing data in GraphPad Prism
  • Running scripts in Rstudio

Our Immuno-Oncology group is looking for a Senior Research Associate or Associate Scientist with deep experience performing preclinical studies in mouse models of cancer. This is a hands-on role in which you will help execute efficacy, tolerability, and PK/PD experiments to support multiple pipeline programs. We are looking for candidates who have the technical skills to instruct research associates and the organizational proficiency to coordinate the logistics for larger experiments.

Primary Responsibilities

  • Perform in vivo efficacy, tolerability and PK/PD studies in a variety of mouse subcutaneous tumor models to support the evaluation and prioritization of drug candidates, including sample banking and data collection
  • Coordinate with scientific study leads and vivarium research technicians to ensure studies go smoothly
  • Report to the Manager of Vivarium Operations and collaboratively establish study goals & timelines
  • Analyze experimental data using MS Office and GraphPad Prism and prepare reports for study leads

Required Qualifications

  • MS or BS in biotechnology, animal science, pharmacology, biology or a related discipline
  • 5+ years of full-time experience working with mouse models of cancer
  • Proficiency with mouse dosing techniques (IV, PO, IP, SC)
  • Experience sampling and processing blood, normal organs, and tumors from mice
  • Detail-oriented and has a track record of organized record-keeping
  • Excellent planning skills and an ability to meet deadlines
  • Strong communication and interpersonal skills

Soteria Biotherapeutics is seeking a highly motivated Research Associate or Senior Research Associate with expertise in antibody purification and characterization to join our team in San Mateo. Soteria is a discovery-stage therapeutics company focused on the development of next-generation T-cell engager therapies. Our proprietary technology, based upon founding research by UCSF professor and renowned protein engineer Jim Wells, enables precise control of T-cell activation and targeting. Primary Responsibilities

  • Contribute as an integral member of an exciting biotherapeutics company
  • Carry out the protein purification of antigens and antibodies of various formats
  • Develop and apply multistep purification routes to isolate complex bispecific antibodies using an AKTA FPLC
  • Characterize the purity and developability (i.e. physical and chemical stability) of antibody therapeutic candidates
  • Work closely with teams of protein engineers, immunologists, and in vivo pharmacologists to optimize protein designs for ideal therapeutic properties
  • Prepare plasmids and perform transient transfections of mammalian expression cell lines for the production of biologics
  • Maintain an accurate and well-organized digital lab notebook and sample inventory
  • Independently design and execute experiments planned by the scientific team
  • Document and summarize experimental results and communicate findings to the scientific team

Qualifications

  • MS or BS in biology, biochemistry, molecular biology, or related discipline
  • At least 2 years of post-graduate experience in an industrial setting
  • Experience with common antibody purification techniques (e.g., Affinity, SEC, HIC, Ion Exchange, etc.) using an AKTA FPLC
  • Knowledge of protein biochemistry and competence in routine analytical protein characterization techniques (SDS/PAGE, DSF, HPLC-SEC, BLI/SPR, etc.)
  • Experience in expression, purification, and characterization of bispecific, multi-specific, and alternative antibody formats
  • Competence with mammalian cell culture and transient transfections is a plus
  • Experience with automation is a plus
  • Demonstrated ability to execute experiments both independently and as part of a team
  • Strong interpersonal, verbal, and written communication skills and a track record of highly organized record keeping

The Senior Research Associate I/II is a highly motivated scientist who will contribute to the advancement of drug candidates in the Soteria Biotherapeutics, Inc. oncology pipeline from discovery research to early development. He/she will use bench laboratory skills in analytical and bioanalytical chemistry, biochemistry, and/or molecular biology to execute a variety of standard and novel methods to support bioanalysis and bioassays as part of the product development pipeline at Soteria Biotherapeutics, Inc. and will utilize and maintain state-of-the-art analytical equipment.

Key Responsibilities

This individual will be responsible for implementation of analytical and bioanalytical assays in support of multiple areas of research and preclinical development, including Bioanalytical Sciences, CMC, and Protein Sciences.  While studies in the labs are executed in a non-regulated non-GxP environment, high-quality standards are expected, and transfer of methods to partner labs is required.  Bioanalytical methods may be used to quantitate therapeutic drug levels, detect and characterize immune responses evoked by drug candidates, measure biomarkers, and characterize drug candidates from buffered solution and various biological fluids and tissues.  This individual will perform bioanalytical measurements, calculations, data entry, and lab notebook maintenance meticulously and with a high level of integrity.  The candidate will contribute data to regulatory filings related to bioanalytical methods and characterization of drug candidates and their biological activities.  Additionally, he/she will present data at weekly science meetings and maintain an open and collaborative relationship with peers.

Knowledge, Experience and Skills

  • A BS or MS in analytical chemistry, biochemistry, immunology, or cell biology or a related field with a minimum of 7 years for BS or 4 years for MS of relevant industry experience in bioanalytical aspects of biopharmaceutical development and analytical characterization of protein therapeutics.
  • Experience in ligand-based assays and protein quantitation bioassays (MSD, ELISA, Luminex, or Alphascreen) is required.
  • Experience with mass spectrometry, specifically intact mass, peptide mapping, and post-translational modification analysis is preferred. Experience with Thermo orbitrap instrumentation and BioPharma Finder software is desirable.
  • Experience with small molecule quantitation by LC-MS/MS is preferred.
  • This individual must have excellent interpersonal and both oral and written communication skills along. Method development experience is required.
  • Experience in immuno-oncology drug development (both small molecule and biologics) is highly desired.
  • Experience in method transfer to contract labs is beneficial.
  • Works well in a small, highly collaborative team setting.
  • Can meet deadlines in a fast-paced work environment.
